The primary aim of our team is to delve into the conserved RNA elements found within coronavirus genomes, with a specific emphasis on their involvement in RNA synthesis. We plan to employ cutting-edge methodologies, including reverse genetics, RNA structure probing, mutational profiling via sequencing, and reporter assays. These techniques will aid in unraveling the functional significance of the conserved RNA elements. Through this comprehensive approach, we aim to understand better the intricate RNA-RNA and RNA-protein interactions that regulate viral RNA synthesis. Such insights are vital for unraveling the mechanisms underlying coronavirus replication.
