Spatio-temporal changes in protein and genetic networks – functional relevance in right heart failure
Pulmonary artery banding in weanling rats will be used to study the transition from compensated (adaptive) right ventricular (RV) hypertrophy to RV failure. By performing pulmonary or aortic artery banding models in weanling rats we found profound functional and transcriptional stage- and ventricle-specific differences in the adaptation of the heart to chronic pressure overload. Bioinformatic analyses implicate deregulated phosphatase networks in this process. By comparative deep molecular phenotyping we will unravel the changes in signal transduction and in chromatin and proteome landscapes upon RV or LV failure and the contribution of deregulated protein networks to right heart pathology. Based on these data we will also explore novel therapeutic targets in cardiac signaling.
