Autism-relevant transcriptional networks in human neural stem cells

Autism spectrum disorders (ASDs) are neurodevelopmental disorders that affect 1% of the population.

Chromatin modifiers and transcription factors are frequently mutated in ASDs. However, the protein and gene networks they function in are not understood. We will biochemically purify ASD-associated factors from human neural stem cells and identify their protein interaction networks by mass spectrometry. Our new method will allow 5-10 fold more interactors to be identified than current standards and will provide quantitative information on their stoichiometry. Subsequently, we will determine common target genes of several selected ASD-related factors to define relevant pathways and diagnostic marker genes. These pathways may ultimately be targeted by future early-intervention therapeutic strategies.
